I generally will be listening to a song, and I'll think of a variation on whatever is in the song. Then I try to replicate it, but I can never get it down right. Generally I try and make it EXACTLY the way I want it, and I'm never able to.

Transcribing lines from other songs helps a lot because it will give you a good idea of what the intervals will be.

Some music theory would definitely help you get ideas from your head to your keyboard, as would some sight-reading/ear training courses. Knowing your scales and modes will especially help you with melodies. My first degree was in theory/composition and, if there's one rule that I learned regarding composition, it's that there are no rules. But, there are some approaches that work well for me:
- get away from your mouse/keyboard - your piano/MIDI controller keyboard is much more conducive to creativity and will let you focus on the melody that you're trying to create, rather than fussing with pencils, erasers, note lengths, grids, etc.
- start with chord progression and improvise over the scales until you find something that works, and build on it; a solid understanding of scales/modes will help you out here,
OR
- start with a rhythm that you like (maybe even borrow one from another song's melody) and come up with your own tune for that rhythm. Then, modify the rhythm if necessary. Add chords to your melody - again, a solid understanding of modes and chord progressions will help.
- listen closely to and study great melodies; pay special attention to the intervals (are they small intervals vs. big leaps, up/down, how it evolves over time, etc.).
