If I play a new place then I like to research it. Check the web, other DJs that have played there and generally get a feel for what people might want and what you can get away with.
I may have a drink then again if I arrive just before the set having played somewhere else about an hour or two ago then I like to stand on the dancefloor for a minute and get a feel of the soundsystem if I have the time to do it, just to adjust my ears a little. Listen for the key of the last track that the warm up DJ is playing and plan a little in my head and then will read how that works once I start playing.
Other than that I don't really prepare for gigs, although I know roughly what I want to be playing in part due to music policies so will work around that.
For the first gig of the night I will usually listen to the guy before me and guage what state he has managed to work the crowd into.
If I'm warming up then I will always start off with something low key that I like, people aren't going to dance that much so you might as well enjoy yourself for the first 30 mins or so.
When it comes to radio then I tend to plan my mixes a bit more. I approach them more as I would a demo as I see them as being pretty much the same thing except one step up from it.
Cheers

^spot on
when i show up for a gig i'll drop my bags off in the booth then grab a drink and stand off to the side of the dancefloor while getting a feeling of what the crowd is feeling and what the dj is laying down
i'm usually pretty quiet and focused at this point, best to just leave me be
like nem said, i'll usually have a good general idea of what i'm going to play, with a few key tracks in mind and a couple good escape plans but i usually don't know what i'm playing until i'm playing
and yes, i like to pre-plan my radio sets a little more since it is in a way like a demo
